Most of the fixatives immobilize the tissue antigens by forming cross-linkages. Sometimes extensive cross-linking of proteins in high concentrations can also lead to the masking or destruction of antigens, which leads to poor staining or no staining at all. This loss of staining can be restored to certain extent by pretreatment of tissue sections with proteolytic enzymes. The unmasking of tissue sections is an important requirement for the optimal staining of antigens like immunoglobulins, Keratins, Carcinoembyonic antigen, Factor VIII-related antigen, Testosterone and to a lesser degree S-100 protein.

The Pronase reagent is a stabilized solution of wide spectrum Pronase in a ready-to-use form.


Specificity:
The Pronase Reagent is a stabilized solution of wide spectrum Pronase in a ready-to-use form. Most fixatives immobilize tissue antigens by forming cross-linkages. Sometimes extensive cross-linking of proteins in high concentrations can also lead to the masking or destruction of antigens, which leads to poor staining or no staining. This loss of staining can be restored to a certain extent by pretreatment of tissue sections with proteolytic enzymes. The unmasking of tissue sections is important for the optimal staining of antigens like immunoglobulins, keratins, carcinoembryonic antigen, factor VIII related antigen, testosterone, and to a lesser degree, S-100 protein.

Storage:
Refrigerate at 4°C. Do not freeze.

Notes:
Procedure: 1. Hydrate the tissue sections in 1X Automation Buffer* for 2 minutes.

2. Drain off the buffer and introduce the Pronase reagent. Make sure the sections are completely covered.

3. Incubate the slides for 5 minutes** at room temperature.

4. Drain off the Pronase reagent and wash the slides with 1X Automation Buffer.

* Biomeda's 10X Automation Buffer (Cat. No. M30).
** Prolonged incubation may result in complete digestion of the tissue and/or it's detachment from the glass slide.

Do not use beyond the expiration dated stated on the label.
